In the last several years, the use of dendritic cells has been studied as a therapeutic strategy against tumors. Dendritic cells can be pulsed with peptides or full-length protein, or they can be transfected with DNA or RNA. However, comparative studies suggest that transfecting dendritic cells with messenger RNA (mRNA) is superior to other antigen-loading techniques in generating immunocompetent dendritic cells. Cytotoxic T lymphocyte-associated antigen-4 (CTLA-4) is a negative regulator of T cell activation, which competes with CD28 for B7.1/B7.2 binding, and which has a greater affinity. Fusion of specific antigens to extracellular domain of CTLA4 represents a promising approach to increase the immunogenicity of DNA vaccines. In this study, we evaluated this interesting approach for CTLA4 enhancement on prostate stem cell antigen (PSCA)-specific immune responses and its anti-tumor effects in a prostate cancer mouse model. DNA vaccine coding a p23 protein of Cryptosporidium parvum fused with Fc portion of immunogloblin G induces higher level of interferon-γ expression in mice OAK

To develop DNA vaccines against Cryptosporidiosis, a plasmid coding an immunodominant protein of Cryptosporidium parvum sporozoite, p23 (pCX-p23) and another plasmid coding a fusion protein containing whole the p23 and the Fc portion of mouse immunogloblin G1 (pCX-p23Fc). Vaccination of BALB/c mice with the plasmid, pCX-p23 and pCX-p23Fc induced the production of antibodies against p23. Although both of splenocytes of mice immunized with the plasmids pCX-p23 and pCX-p23Fc expressed interleukin-4 and interferon-8, after the in vitro stimulation by p23 antigen, the interferon-8 expression level of pCX-p23Fc immunized mice was much higher than that of pCX-p23 immunized mice. The neonatal diarrhea in swine caused by enterotoxigenic Escherichia coli (ETEC) is responsible for high mortality and low growth rate in pigs and it is mainly dependent on the capacity of E. coli to attach to the surface of the small intestine, a property mediated by fimbria. In this study the faeC gene, which codes for the minor fimbrial subunit of E. coli K88ab, was cloned in the eukaryotic expression vector pcDNA3, associated or not to the Kozak sequence. The use of specific combinations of antigens and adjuvant represents a promising approach for increasing the immunogenicity of DNA vaccines. In the present study, we evaluated the immunity and antitumor effects of DNA vaccines with G250 as the target antigen in a mouse model of renal cell carcinoma. Genetically attenuated microorganisms, pathogens, and some commensal bacteria can be engineered to deliver recombinant heterologous antigens to stimulate the host immune system, while still offering good levels of safety. A key feature of these live vectors is their capacity to stimulate mucosal as well as humoral and/or cellular systemic immunity. This enables the use of different forms of vaccination to prevent pathogen colonization of mucosal tissues, the front door for many infectious agents. The induction of systemic (IgG) and mucosal (IgA) antibody responses against the colonization factor I antigen (CFA/I) of enterotoxigenic Escherichia coli (ETEC) was evaluated in mice primed with an intramuscularly delivered CFA/I-encoding DNA vaccine followed by two oral immunizations with a live recombinant Salmonella typhimurium vaccine strain expressing the ETEC antigen. The booster effect induced by the oral immunization was detected two weeks and one year after the administration of the DNA vaccine. Serological responses in sheep injected with plasmids encoding bovine herpesvirus 1 (BHV-1) gD glycoprotein Arq. Bras. Med. Vet. Zootec.

A genetic vaccine consisting of the bovine herpesvirus-1.2a (BHV-1.2a) glycoprotein D (gD) gene under the control of the cytomegalovirus immediate-early promoter/enhancer was generated and administered to sheep intramuscularly in the neck. All animals developed serum antibodies which recognized the homologous antigen (BHV-1.2a strain BH-83) and also exhibited cross-reactivity against the heterologous antigen (BHV-5 strain EVI-190). Three intramuscularly injections were given but serological responses were not improved after the second inoculation. Specific antibodies were detected against BHV-1.2a until at least 12 months after the first inoculation. However, the capacity to induce antibodies against BHV-5 was lower and of shorter duration than to BHV-1.2a.

ABSTRACT DNA vaccines have been shown to be an effective approach to induce antigen-specific cellular and humoral immunity. However, the inability of DNA vaccines to elicit strong immune responses in clinical trials limits the application of DNA vaccines.
